Slow Computer, General Check Up

Customer's Description:

The first problem is that I can't run defrag'. It says that Chkdsk/f is scheduled to run at the next start, but it doesn't. Another message at start-up says that the volume cannot be accessed. I have tried running chkdsk/f from the command prompt but I just get the message saying that it will be run at the next start-up. Overall the computer is running very slowly. I have PC-Cillin running and up to date plus Spyware Doctor and Registry checker.
